BURG, MI., L. STONER, J. CARDINAL, AND N. GREEN. Furosemide effect on isolated perfused tubules. Am. J. Physiol. 225(l): 119-124. 1973 .-Proximal convoluted tubules, thick ascending limbs of Henle’s loop, and cortical collecting tubules were dissected from rabbit kidneys, and perfused in vitro. The effect of the diuretic agent furosemide on electrolyte transport and electrical potential difference (PD) in these segments was determined. Furosemide in the lumen (lo-” to lOA M) inhibited active Cl transport in the thick ascending limbs of Henle’s loop with resultant decrease in net NaCl absorption and electrical PD. Concentrations of furosemide as high as 10 -4 M in the bath had little effect. Furosemide at low4 M also had no definite effect on proximal convoluted tubules and cortical collecting tubules whether placed in the bath and/or the lumen. We conclude that the principal renal action of this drug is to inhibit active Cl transport in the thick ascending limb of Henle’s loop.

Perfusion of isolated tubules of the shark rectal gland. Electrical characteristics and response to hormones.
Both the mammalian thick ascending limb of Henle's loop and the shark rectal gland actively transport Cl against an electrochemical gradient by mechanisms involving hormone-sensitive NaCl transport. In contrast to mammalian renal tubules, individual tubules of the shark rectal gland previously have not been perfused in vitro. Using a combination of renal slice and microdissection techniques we ...
متن کاملK(+) transport in the mesonephric collecting duct system of the toad Bufo bufo: microelectrode recordings from isolated and perfused tubules.
We studied the mechanisms of K(+) transport in cells from isolated and perfused collecting tubules and ducts from the mesonephric kidney of the toad Bufo bufo. Cells were impaled with microelectrodes across the basal cell membrane. The basolateral membrane potential (V(bl)) depolarized upon change of bath [K(+)] from 3 to 20 mmol l(-1) demonstrating a large K(+) conductance in this membrane. In...
متن کاملSalidiuretic action by genistein in the isolated, perfused rat kidney.
The urinary isoflavonoid genistein inhibits membrane Na-K-Cl cotransporters at similar concentrations as furosemide, but the significance of this action is unknown. Genistein was therefore investigated in rats for its potential salidiuretic actions. In the isolated, perfused rat kidney, genistein induced a maximal salidiuretic action similar to that of furosemide but was 3 to 5 times less poten...
متن کاملCardiac effects of piretanide and furosemide on intact anesthetized dogs and on isolated atria.
The effects of piretanide and furosemide on systemic arterial blood pressure and heart rate were examined in the anesthetized dog and the effects on atrial rate and contractile force were assessed in isolated atrial muscle perfused with heparinized arterial blood from a donor dog. When piretanide was administered intravenously to intact dogs, the depressor and bradycardic responses were produce...
متن کاملEffects of anion-transport inhibitors on NaCl reabsorption in the rat superficial proximal convoluted tubule.
The effects of anion-transport inhibitors on volume reabsorption, and total CO(2) concentrations were examined by in vivo microperfusion of superficial proximal convoluted tubules of rats. The luminal perfusion solution was a high-chloride, low-bicarbonate solution like that in the in vivo late proximal tubule. The anion-transport inhibitors were only added to the luminal perfusion solutions. I...
